Tilton began her acting career in 1976 appearing on an episode of "Happy Days". She then had a role in the movie "Freaky Friday" with Jodie Foster and then appeared in an episode of "Eight is Enough" in 1977. In 1978 she starred in two movies, "Sweater Girls" and "Big Wednesday".
It was then in 1978 that she starred in "Dallas" as the Grandaughter of Jock Ewing played by Jim Davis. She also co-starred with Larry Hagman (J. R. Ewing), Linda Gray (Sue Ellen Ewing), Patrick Duffy (Bobby Ewing), Steve Kanaly (Raby Krebbs), Barbara Bel Geddes, and Victoria Principal (Pamela Barnes Ewing). She took some time off from "Dallas" in 1986-87 and returned the following season and stayed for three more years. During the "Dallas" years Charlene also appeared in other TV shows and movies such as "Diary of a Teenage Hitchhiker" (1979), spin-off show "Knots Landing" with Ted Shackleford and Joan Van Ark (1980), "Laverne and Shirley" and "Fantasy Island" with Ricardo Montalban and Hervé Villechaize (1980), "The Fall of the House of Usher" (1982), "The Love Boat" with Gavin MacLeod (1980-87), and "Murder, She Wrote" (1987).

After "Dallas" Charlene starred in some feature films such as "For Parents Only" (1991), "Problem Child 2" (1991), "Deadly Bet" (1992), "The Silence of the Hams" (1994), "Totally Blonde" (2001), and "Superhero Movie" (2008). There were also some made for TV movies in there as well such as "Star Witness" (1995), "Bar Hopping" (2000), "A Distant Thunder" (2005), and "Point of Entry" (2007).

Charlene continues to entertain as she has appeared in three movies in 2010 including "Mean Parents Suck" (2010), "Paranormal Calamity" (2010), and "#1 Cheerleader Camp" (video only 2010).
In 2012, Charlene revived her role as Lucy Ewing in the new TV version of Dallas co-starring Josh Henderson (John Ross Ewing), Jesse Metcalfe (Christopher Ewing), Jordana Brewster (Elena Ramos), Brenda Strong (Ann Ewing), Julie Gonzalo (Rebecca Sutter), Patrick Duffy (Bobby Ewing), Linda Gray - Sue Ellen Ewing, and Larry Hagman (J. R. Ewing).

Also in 2012, Tilton starred in the film Edge of Salvation with Jeremy London, Jonathan Lipnicki, and Kelly Washington; Child of the '70s starring Geri Jewell, Bruce Vilanch, Donna Pescow, and Sebastian La Cause; and Buttwhistle co-starring Trevor Morgan, Elizabeth Rice, Stella Maeve, and Analeigh Tipton.
